A joke like that can feel jarring when the moment around it is mundane. Whether it ruins the mood probably depends on how much crude humor you enjoy. If it’s a one-off for you, it may be easy to shrug off. If that tone keeps coming up, I can see it changing how you feel about the companion.

One awkward line wouldn’t necessarily put me off a character, but the contrast can make the writing feel less natural. I’d judge it by whether the companion has other dialogue that works for me, rather than letting one joke define the whole relationship.
