Is it possible to go back and replay a mission from the main story?

It is possible to replay then but not right away. Once you finish the main campaign, which can be done in 4-6 hours (maybe less with a good strike team), you can speak with Ikora and she will allow you to “meditate” on your journey, which allows you to replay past missions.
